How many DPO should I take a pregnancy test?

The most reliable day is 14 DPO (the typical missed period) or later. Testing before 12 DPO produces many false negatives because implantation occurs 6-12 DPO and hCG needs 1-2 days after implantation to become detectable.

What does a negative test at 10 DPO mean?

Very little. Many healthy pregnancies have not yet produced detectable hCG at 10 DPO, especially those that implanted on day 9 or later. Retest at 12-14 DPO with first-morning urine.

It is built directly from the implantation evidence: implantation occurs 6-12 days after ovulation, most often days 8-10 (Wilcox 1999), hCG appears in maternal blood only after implantation, and even a perfect test detects at most 90% of pregnancies on the first day of the missed period because 10% have not yet implanted (Wilcox, JAMA 2001). In modern daily-urine cohorts, only ~5% of newly pregnant women had detectable urinary hCG on day 8 after the LH surge, rising to 43% on day 9 (Johnson 2009). The asymmetry to remember: an early positive is usually meaningful; an early negative means very little. If the answer matters this week, a quantitative blood test beats any strip.
